Comments: We have generated transgenic mouse lines using the liver-specific regulatory signals of the transthyretin gene. Two lines have been established that show different expression patterns in a test system involving a cross with the mice carrying a loxP-flanked gene. The TTR19-1 transgenic line showed a liver-specific recombination of the loxP sites with an efficiency of 50 %, resulting in production of somatic mosaicism exclusively in liver. The other line, TTR10-3, demonstrated recombination more broadly in digestive organs such as liver, pancreas, gut, and gallbladder with efficiencies of 95%, 100%, 50% and 50 %, respectively.

Comments: Deletion in many tissues including the female germline \(Line A\). This line was used to generate null alleles from a floxed locus \(in press\). Recently, we have isolated one line which shows no recombination in the male and female germline \(Line D, unpublished\). All MMTV-Cre lines we have generated show excision not only in the mammary gland but also in a lot of other tissues. However, embryonic lethality has never been observed after deletion of genes reported to be lethal in complete null mice suggesting a mosaic expression pattern of MMTV-Cre in those tissues.

Notes: We have injected the following construct\: tetO\>rtTA--Cre\tetO, i.e. both rtTA and Cre are under the control of tetO. We speculated that a light leakage of the promoter will be enough to "prime" the enhancement following Dox administration. We also add NRSF sites following the two promoter sites to try to reach a certain specificity of expression in the brain. 6 lines have been obtained, and are currently under investigation.
